Alargenumberofphysiologicalandpathologicaleventsleadtosocalledstressconditionstowhichtheorganismcanadaptwithincertainlimitsbyregulatingtheactivityofaseriesofenzymaticcascades.Moststressresponsesarecharacterizedbyanincreasedgenerationof“freerADIcals”whichencompassbothreactiveoxygen(ROS)andnitrogenspecies(RNS).Mostofthesespeciesreactwithmacromoleculesoftheorganism,i.e.aminoacids,proteins,lipidsandDNA,leadingtofunctionalalterationswhichcaneitherparticipatetoadaptationorleadtocelldeath.
Oxidativestresshowever,hasdifferentfunctionalconsequencesdependingnotonlyuponitsintensityandduration,butalsouponthenatureofthefreeradicals,ROSorRNS,generated.Interestingly,thenatureofthesereactivespeciesdependsontheratiobetweentheinitiallyproducedROS,superoxideanion(O2.-),andnitricoxide(NO).Indeed,O2.-veryrapidlyreactswithNOandtherefore,aslongastheNO/O2.-ratiois=1,O2.-willthereforepreferentiallyreactwithNOratherthanwithmacromolecules,thusgeneratingreactivenitrogenspecies(RNS):nitrosonium(NO+),N2O3andperoxinitrite(ONOO-).TheseRNSinduceposttranslationalmodifications:forNO+andN2O3,S-nitrosation(Cys-SNO)andforONOO-tyrosinenitration(Tyr-NO2),methioninesulfoxidation(Met-SOH)andthioloxidation(RS-S-R).WhentheO2./NOratiobecomes>1,theO2.-andNO.2ionsandthereaftertheOH.radicalscauseirreversIBLeoxidationsandperoxidationsofmacromoleculeswhichgenerallyleadtocelldeath(1-4).ThemonoclonalantibodieshavebeenselectedfortheirhighaffinityandspecificitytowardsTyr-NO2residues.Theydonotcross-reactwithTyrorTyrderivativessuchasaminotyrosine,chlorotyrosineorphosphotyrosine,neitherwithnitroTrpwhichcanalsobegeneratedinresponsetoperoxynitrite.TheyalsorecognizeTyr-NO2residuesinvarioussequencesasshownbytheirABIlitytorecognizenitratedproteinsincludingalbumin,ovalbumin,insulin,hemoglobin,KLHandvariouscytoplasmicandmitochondrialproteins.
